Assertion : All expression vectors are c...

Assertion : All expression vectors are cloning vectors and vica versa.
Reason : Expression vectors have at least the regulatory sequences i.e., promotes, operators, ribosomal binding sites, etc having optimum function in the chosen control but not origin of replication.

The correct Answer is:
D

All vectors used for propagation of DNA insertes in a suitable host are called cloning vectors. But when a vecor is designed for the expression i.e, production of the protein. Specified by the DNA insert, it is termed as expression vector. Expression vectors need regulatory machinery suitable for the host, e.g., if an eukaryotic gene is to be expressed in prokaryotes, the expression vectors should contain gene without introns because prokaryotes lack introns removal mechinery for transcripts. But all vectors, including expression vectors must contain 'origin of replication' so that they can replicate when the host cell divides and their presence is ensured in all progeny.

Assertion : Restriction enzyme cuts both vector DNA and foreign DNA at the same site Reason: Both plasmid and bacteriophages have the ability to replicate within bacterial cells independent of the control of chromosomal DNA

The different steps of recombinant DNA technology are given below randomly. (i) Isolation of the DNA fragments or genes to be cloned (ii) Introducting of the recombinant DNA into a suitable cell (usually E. coil) called host (transformation) (iii) Multiplication/expression of the introduced gene in the host (iv) Selection of the transformed host cells, and identification of the clone containing the desired gene/DNA fragment (v) Insertion of the isolated gene in a suitable plasmid vector Which of the following represents the correct sequences of steps ?

(II) Competent bacterial cells cannot take up foreign DNA. (III) In elution ,the separated bands of DNA are cut out fron agarose gel and extracted from the gel piece. (IV) The downstream processing and quality control testing vary from product to product .

(b) E coil cloning vector pBR322 have several restriction sites, Ori, antibiotic resistance genes & ROP. (c ) The downstream processing and quality control testing vary from product to product. (d) Competent bacterial cells cannot take up the plasmid.

Read the following statement regarding DNA recombinant technology. Identify the statements that are true and false and select the appropriate option. I. Selectable marker helps in identifying and eliminating non-transformants and selectively permitting the growth of the transformants. II. Plasmids have the ability to replicate within bacterial cells independent of the control of chromosomal DNA making it a suitable cloning vector. III. The DNA fragments for isolation can be separated by a technique known as thin-layer chromatography. IV. A technique where cells are bombarded with high velocity micro-particles of Au or W coated with DNA is known as micro-injection. {:(,I,II,III,IV),(1,"False","False","True","True"),(2,"True","True","False","False"),(3,"True","True","True","True"),(4,"False","False","True","False"):}
