Assertion : E.coli having pBR322 with DNA insert at BamHI site cannot grow in medium containing tetracyline.
Reason : Recoginition site for BamHI is present in `ter^(R)` region of pBR22.

Text Solution

Verified by Experts

The correct Answer is:
A

pBR322 has recognition sites for several commonly used restriction enzymes. Recognition site for BamHl is present in `tet^(r)` region i.e., region responsible for tetracyline resistance. When an insert is added at the BamHl recongnition site the gene for tetracycline resistance becomes non-functional and the recombinant bacteria with plasmid `pBR322` that has `DNA` insert at BamHl lose tetracycline resistance.

Assertion: E. coli having pBR322 with DNA insert at BamHI site cannot grow in medium containing tetracycline. Reason: Recognition site for Bam HI is present in "tet"^(R) region of pBR322.

Draw a schematic diagram of the E. coli cloning vector Pbr322 and mark the following in it : (a) ori (b) rop (c) ampicillin resistance gene (d) tetracycline resistance gene (e) restriction site BamHI (f) restriction site EcoRI

Assertion (A): Any fragment of DNA, when linked to the ori region, can be initiated to replicate. Reason (R) : Ori is a genetic sequence that acts as the initiation site for replication of DNA.

E. coli cloning vector pBR322 showing restriction sites ori and antibiotic resistance genes is given below. Select the correct option .

Some foreign DNA fragment is attached to Cla I site of pBR 322. This recombinant vector in used to transform Escherichia coli. Then on two different media, one containing ampicillin and the other containing tetracycline. The transformed cells containing the recombinant vecotr will

The vector also have one unique recognition site to enable foreign DNA to be inserted into the vector during the generation of an rDNA molecule. Most of the commonly used vectors contains unique recognition sites region of DNA which is referred to as a polylinker or multiple cloning site (MCS). An MCS provides:

Assertion : Origin of replication (ori) is a definite region in E. coli DNA where the replication originates. Reason : The vectose in r-DNA procedures provide the origin of replication.

If the gene of interest is inserted at the BamHI site in pBR322, the recombinant plasmid will
