Explanation

Colchicine is called artificial polyploidizing agnet. It is used for chromosomal doubling. A haploid cell can be converted to diploid cell and thereafter tetraploid cell by colchicine treatment. A callus produced by pollen culture is haploid. If it is treated with colchicine diploid callus is produced. This diploid allus will then be converted to tetraploid callus or octoploid callus by further givein treatment of colchicine while growing on culture medium. Thus the ploidy level of callus can be converted from haploid to diploid then to tetraploid or octaploid, as per need. All such calli will carry homozygous character (pure line).
