What procedure do you follow to understand the effect of plant growth hormones (in agar medium) in the terminal portion of the tip of stem (coleoptile) ?

Explanation

Required material for observation of Rizopus in the lab : microscope, roti (or) bread , plastic cover , water , slide, coverslip, etc.
Preparation :
(1) Take bread (or) Roti to grow Rizopus in a controlled environment.
(2) Keep it in plastic bag and sprinle water on it . It absorbs water . And make it sure some amount of air is inside the bag and seal the bag .
(3) Keep it in a distant place from other food sources. Ensure that dark and warm conditions are maintained . Then Rhizopus starts growing within two (or) three days .
Observations :
(1) Take a slide and put water drop on the niddle with the help of tooth pick collect Rizopus and keep it on the middle of the slide .
(2) Keep cover slip on the slide , ensure that water bubbles are not there.
(3) Remove excess water on the sides of coverslip with the help of Tissue paper . Then observe it under the microscope.
Precautions :
(1) Maintain some distance between Rizopus plastic bag and other food sources.
(2) Wash your hands properly if you touch the Rhizopus with direct hand .
