Gene Amlification. (Polymerase chain Reaction or PCR). It is a technique of in vitro repeated duplication of selected intact or broken DNA. Million of copies of selected DNA can be made within or few hours. It requires sample DNA, primers, nucleotides and polymerases. Thermostable DNA polymerase or Taq polymerase of thermophilic bacterium Thermus aquaticus is used. The mixture is heated (for separation of two DNA starands ) and cooled (for DNA replication) alternatively. The whole cycle takes 30 minutes. The process is repeacted a few times. By using this technique, DNA of a dead man was amplified and analysed to find HIV in his cells.
