Why is only rubber used to make cycle seat instead of wood or iron ?

Because, every atoms or molecules of rubber are bonded together by intermolecular forces and remains in a stable equilibrium. When cycle is running and passes from the pit-hill type ground the material of its seat experience deformation and their atoms or molecules displaced from its position but when the bicycle is at rest, intermolecular forces placed them in original position. Hence deformation does not occurs and no shocks will observed. This is not in the case of wood or iron.
