Steps for isolation of DNA-
The bacterial cells/plant or animal tissue is treated with enzymes such as lysozyme (bacteria)/ cellulase (plant cells), chitinase (fungus) to break open the cell releasing DNA along with other macro molecules such as RNA, proteins, polysaccharides and also lipids.
The RNA can be removed by treatment with ribonuclease, proteins by treatment with proteases and others by appropriate treatment respectively.
Purified DNA ultimately precipitates out after the addition of chilled ethanol
DNA that separates out as a collection of fine threads in the suspension can be removed by spooling.
