How is a mature, functional insulin hormone different from its prohormone forms?

In human, insulin is synthesised as a prohormone or proinsulin which contains three chains A, B and C (stretches of peptides). This C peptide is removed during maturation of insulin, while A and B chains are linked together by disulphide bridges.
