Safranin appears a brilliant red in chromatin, nu- clei, lignified and suberised or cutinised cell wall. Fast green appears as brilliant green in cytoplasm and cellu- losic cell walls. Thus, lignified tissue such as xylem and scleren-chyme tum red and Parenchyma, Collenchyma and Phloem remain colourless on staining with safranin. When double stained with fast green then the colourless ones now turn green. Thus xylem primarily appears red and phloem primarily appears green.
