Selection of recombinants due to inactivation of antibiotics is cumbersome as it requires simultaneous plating of two plates having different antibiotics, Where as marker gene coding for chromogenic substrate differentiate recombinants from non-recombinants on basis of ability to produce colour in presence of chromogenic substrate.
