Addition of nucleotides can be done on a pre synthesised DNA strand with 8-10 base pairs of DNA, i.e., primer. PCR is the technique where double-stranded DNA is taken and converted into single strand and both the strands are used as a template. Primers are used for the synthesis of new strand of DNA. If primers which are used are self-complimentary then they will not be available for the extension.
