DNA finger printing technology is a technique introduced by Alex Jeffry. It is mainly based on satellites like (VNTR) - Variable number of tandem repeaters.
Steps involved in DNA fingerprinting technology are as follows:
(i) Collection of sample : Any parts of the body be it be hair follicle saliva, semen blood can be taken as a sample
(ii) Separation of DNA : The sample has to be with celluloses, pectionases, lysozymes to get it from the cell wall. If the obtain DNA is very small it can be amplified by PCR.
(iii) Fragmentation and Agarose get Electrophoresis: The obtained DNA can be fragmented suitable REN.s and the fragments are according to their sizes in get electrophorus.
(iv) Obtaining single stranded DNA : The gel plate is treated with alkali to obtain single strands of DNA.
(v) Southern Blotting: The get plate is transferred to the micro cellulose paper, this technique is called southern Blotting.
(vi) Hybridizing with probe DNA : The single stranded DNA is hybridized with radioactive DNA.
(vii) Autoradiography : The X-Ray film is placed over the nitrocellulose sheet and then it is auto radio graphed to obtain large number of bands of various sizes.
(viii) Comparison of band patterns : The suspected individuals band patterns can be compared with the sample and thus the criminal can be found out.
