Explain the contribution of thermus aquaticus in the amplification of a gene of interest

Text Solution

Verified by Experts

Amplification of a gene is carried out by the tec1mique called Polymerase Chain Reaction to synthesise multiple copies of the desired gene. This tec1mique is based on annealing at `40^@ C - 60^@ C` and synthesis of new strand, i.e., extension and denaturation at high temperature, i.e., `70^@ C - 90^@C` . High temperature inactivates the DNA polymerose. Therefore, a thermostable (i.e., heat stable) Taq DNA polymerase extracted from heat stable bacterium Thermus aquatjeus is used to carry out the process of amplification without any difficulty. It does not require adding of new enzyme to the PCR process.
