(1) Restriction endonucleases cut the DNA fragment at specific site. The site at which there is a cut is called recognition site or restriction site.
(2) Restriction endonucleases have the ability to recognize DNA with specific 4 to 8 base pair sequences. RENs cleave these sequences.
(3) Every restriction site is identified by a specific restriction endonuclease.
(4) At the restriction sites both the DNA strands are cut at the same time at these specific points.
(5) Most of the restriction sites have palindromic DNA and hence called palindromes.
