During genetic engineering experiments r...

During genetic engineering experiments recombinants are distinguished from nonrecombinants by different methods . One of the methods is know as insertional inactivation . Explain the process.

Text Solution

Verified by Experts

In this process a recombinant DNA is inserted within the coding se-quence of an enzyme `alpha` - galactosidase. This results intothe inactivation of the enzyme, which is referred to as insertional inactivation. Presence ininsert results into insertional inactivation of the `alpha` - galactosidase and the colonies do not produce any colour, these are identified as recombi-nant colonies .

Fill up the blanks in the following paragraph by selecting the correct option A. overset("i")("___") methods work on the principle of avoiding chances of ovum and sperms meeting B. overset("ii")("___") is one such method in which the couples avoid coitus from day 10 to 17 of the menstrual cycle C. overset("iii")("___") is another method in which the male partner withdraws his penis from the vagina just before ejaculation so as to avoid insemination D. overset("iv")("___") method is based on the fact that ovulation and therefore the cycle do not occur during the period of intense loctation following parturition.

Assertion : Downstream processing is generally considered more difficult and costlier in plants than that in microbes. Reason : Rhizosecretion is used as a method to facilitate easier recovery of recombinant proteins from plants.

An experiment is performed to measure the molar heat capacity of a gas at constant pressure using Regnault's method. The gas is initially contained in a cubical reservoir of size 40 cm xx 40 cm xx 40 cm xx at 600 kPa at 27^0C . A part of the gas is brought out, heated to 100^0C and is passed through a calorimeter at constant pressure. The water equivalent of the calorimeter and its contents increases from 20^0C to 30^0C during the experiment and the pressure in the reservoir decresases to 525 kPa . Specific heat capacity of water = 4200 J kg^(-1) K^(1). Calculate the molar heat capacity Cp from these data.
